RESUMEN

The chemical composition of Vitex agnus-castus L. (Verbenaceae family) fruits, collected from two regions in Bulgaria (south-central and north-east Bulgaria), was investigated. The content of proteins (5.3-7.4%), carbohydrates (73.9-78.8%), fiber (47.2-49.9%), ash (2.5-3.0%), essential oils (0.5%), and vegetable oil (3.8-5.0%) were identified in the fruits. The composition of the essential oils (EOs) of Vitex fruits from both regions was determined; the main compounds were 1,8-cineole (16.9-18.8%), α-pinene (7.2-16.6%), sabinene (6.7-14.5%), and bicyclogermacrene (7.3-9.0%), but significant differences in the quantitative and qualitative composition of EOs between the regions were found. The EOs of plants from north-east Bulgaria demonstrated antimicrobial activity against the pathogenic species Salmonella abony, Staphylococcus aureus, and Bacillus subtilis, but the Gram-negative bacteria Esсherichia coli and Pseudomonas aeruginosa exhibited resistance to the oil. Linoleic acid predominated in vegetable oil from both regions, followed by oleic acid. ß-sitosterol and γ-tocopherol were the main components in the sterol and tocopherol fraction of the lipids. Phosphatidic acids were the main components in the vegetable oil from north-east Bulgaria, while in the vegetable oil from south-central Bulgaria, all phospholipids were found in almost the same quantity. Overall, significant differences were observed in the chemical composition (proteins, carbohydrates, ash and moisture) of the fruits from the two regions of Bulgaria, as well as in the content of the main components of their essential and vegetable oils.

BACKGROUND: Sildenafil is used to treat erectile dysfunction and pulmonary arterial hypertension and is metabolized in the liver mainly by CYP3A4, thus co-administration with drugs or herbal extracts that affect CYP3A4 activity may lead to drug-drug or drug-herb interactions, respectively. The aim of the present study was to evaluate the influence of single and multiple oral doses of methylxanthine fraction, isolated from Bancha green tea leaves on the pharmacokinetics of sildenafil in rats. METHODS: Rats were given sildenafil alone as well as simultaneously with methylxanthines or ketoconazole. The plasma concentrations of sildenafil were measured with high-performance liquid chromatography method with ultraviolet detection. The pharmacokinetic parameters of sildenafil were calculated by non-compartmental analysis. RESULTS: Concomitant use of sildenafil with a single oral dose of methylxanthines resulted in a decrease in Cmax (p > 0.05), AUC0-t (p < 0.05) and AUC0-inf (p < 0.05), while the administration of sildenafil after methylxanthines pretreatment resulted in an increase in Cmax (p < 0.0001), AUC0-t (p < 0.0001) and AUC0-inf (p < 0.001) compared to the sildenafil group. After co-administration of sildenafil and ketoconazole, a significant increase in Cmax, AUC0-t and AUC0-inf was observed in both of the experiments. CONCLUSION: Drug-herb interactions were observed when sildenafil was co-administered with Bancha methylxanthines in rats. Further in vivo studies about the potential drug interactions between sildenafil and methylxanthines, especially caffeine, are needed to clarify mechanisms underlying the observed changes in sildenafil pharmacokinetics.

Methylxanthines, purine alkaloids found in plants, are found in beverages (coffee, tea, cocoa) and foods (chocolate and other cocoa-containing foods) commonly consumed worldwide. Members of this family include caffeine, theophylline and theobromine. Methylxanthines have a variety of pharmacological effects, and caffeine and theophylline are used as pharmaceuticals. Methylxanthines are metabolized in the liver predominantly by the enzyme CYP1A2. Their co-administration with CYP1A2 inhibitors may lead to pharmacokinetic interactions. Little is known about the possible drug interactions between caffeine and substrates of other CYP450 enzymes. In our study, methylxanthine fractions inhibited CYP3A4 in a concentration-dependent manner. Concomitant consumption of green tea with CYP3A4 substrates could increase the possibility of interactions, and this requires further clarification. The inhibition of CYP3A4 is not only due to the presence of catechin derivatives but methylxanthines may also contribute to this effect.

BACKGROUND: Plant species from genus Carduus are widely distributed in the world and represented in Bulgaria by 14 species. Previous investigations on this genus demonstrated a strong antioxidant potential of extract from some Bulgarian Carduus species. OBJECTIVE: The present study investigates the phenolic profile and the antioxidant potential of different extracts obtained from four endemic Compositae herbs, growing wild in Bulgaria: Carduus armatus Boiss and Heldr., Carduus candicans Waldst. et Kit ssp. globifer (Velen.) Kazmi., Carduus rhodopaeus Velen. and Carduus thracicus (Velen.) Hayek. MATERIALS AND METHODS: Antioxidant capacity of the obtained extracts was estimated with 2,2-diphenyl-1-picrylhydrazyl, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid), and ferric reducing antioxidant power and copper reduction antioxidant assays. Phenolic profile was estimated by high performance liquid chromatography. RESULTS: Eleven phenolic acids and eight flavonoids were quantified in the inflorescences. Sinapic (2760.72 ± 15.68 µg/g dry weight [dw]), chlorogenic (2564.50 ± 19.73 µg/g dw) and ferulic acids (1648.71 ± 19.57 µg/g dw), as well as luteolin (2345.45 ± 18.61 µg/g dw) and apigenin (1332.75 ± 12.05 µg/g dw) were found to be the predominant compounds. The above contents are the highest values found in C. candicans ssp. globifer. The highest established antioxidant activity (AOA) was in favor of the ethanolic extracts, and the extract of C. rhodopaeus affirmed with the highest AOA among the investigated plant species. CONCLUSION: All identified phenolic compounds were reported for the 1(st) time in the studied endemic Carduus species, as well as their antioxidant capacities. The present study revealed that these plant species could be used as sources of antioxidants with potential medicinal properties. SUMMARY: Phenolic acids and flavonoid profiles of four endemic compositae herbs, growing wild in Bulgaria: Carduus armatus, Carduus candicans ssp. globifer, Carduus rhodopaeus and Carduus thracicus, were quantified for the first time by high performance liquid chromatography (HPLC). Eleven phenolic acids and eight flavonoids were determined in the inflorescences. Sinapic, chlorogenic and ferulic acids, as well as luteolin and apigenin were found to be the predominant compounds. The highest values of established phenolic compounds were found in C. candicans ssp. globifer. The studied plant extracts of Carduus species possessed antioxidant activity in favor of C. rhodopaeus and results confirmed 70 % ethanol as more appropriate solvent. The present study revealed that these plant species could be used as sources of antioxidants with potential medicinal properties.
